The question about the best Adsense unit is asked many times in webmaster forums each day and users will definitely get different answers depending on the person and his websites. Certain trends suggest that the medium and large rectangle are the best performing ad units when placed next to content while the leaderboard ad performs best in places where users would look for navigation. I encountered a new layout for the Adsense 468×60 ad units that Google seems to test at the moment. Normally the text Google appears in the right lower corner of the ad unit but on this ad unit it appeared on the upper left. This will, in my opinion, severely reduce the click through rate of that ad unit especially if you place it in your content and not in some place on the top of the page.
